There are some really good ideas above... more variety of flat rides would be great. Games are a part of every park, I'd like to see some in RCT2, also.
A change that would take so much frustration out of building, I think, would be this: you should be able to toggle on an "automatic tree removal" which would (you guessed it) automatically remove trees which are in the path of coasters or paths that you are building. I just don't see the need to right click on 200 trees to build a coaster in the jungle or forest scenarios.
A variation on that also could be an automatic land editor which would eliminate the "Raise or Lower Land" pop-up by digging the tunnels or trenches automatically when going above or below ground with tracks.

I want all the things mentioned above, but here are the big ones I want.
-more flat rides (of course)
-smarter and different looking peeps
-realistic coaster stations (with air gates, etc.)
-realistic looking shops
-theaters for live shows
-inversions on diagonal pieces of track
-a new editor for more complicated tracks
-more landscaping options
-more realistic physics on coasters
-more real parks (US parks, not only large parks like CP, but smaller parks like Worlds of Fun, Holiday World, and Knobels')
I could do without the ride-your-own ride in 3D options. I like RCT's style and if you want to ride your rides, get NoLimits.

The one thing i would love is if you pick a company, or type of coaster, highlight some land and find out if a coaster of that kind could fit, and if it could, then the company would tell you the cost, and desighn and start building!

One of the things that I would love to see, and would be pretty easy to add, is the ability to add realistic tunnels at any point on a coaster (or any tracked ride for that matter). The only way to do it now is raise the land, except on the water slide. When you build a water slide, there’s that option for open or enclosed track. Why not add that type of option when building coasters?
Other than that, pretty much what everyone else has said, with a ton more flat rides being my number one wish.

Several items I would like to see have to do with the stations of several rides:
Split-platform stations for coasters (ala Generation 2 Flying Dutchmen)
Turntable stations for water rides (Most rapids rides I've seen use this, and I've also seen them for flume rides).
Also, for the coasters, banked drops and the ability to make ride elements such as loops and helixs in positions other than 90 degree angles.

I would like to see more realistic breakdown situations on the coasters. For example, have sometypical errors/shutdown... low air pressure, brake slipping, and so on. The coasters running into each other was really unrealistic and annoying. As others have said, it would be cool to see some reducation brakes and other block brakes.
